(b) Insert the rubber tube from the aspirator into the mouth
of the mobile phase container.
(c) Turn ON the power switch of the ultrasonic washer.
(d) Reduce pressure by "opening" the tap water spigot
connected with the aspirator.
(e) As degassing progresses, small bubbles are
discharged and they gradually become larger.
Continue degassing until bubbles are no longer
discharged. This process takes about 5 minutes.
(Refer to NOTE below.)
(f)
After confirming that no bubbles are discharged,
remove the rubber tube of aspirator from the mobile
phase container.
(g) "Close" the tap water spigot.
(h) Turn OFF the power switch of the ultrasonic washer.
(i)
Take out the mobile phase container from the
ultrasonic washer and wipe off moisture adequately.
Then set the container in a liquid chromatograph.
NOTE: Discharge of bubbles varies with the composition
of mobile phase. In case of a mixed mobile
phase with organic solvent (e.g., ethanol 20% +
water 80%), the composition ratio of mobile phase
may vary with degassing time (because the
boiling point of mobile phase differs at a reduced
pressure.) Therefore, degassing time should be
kept constant.
